Molecular evolution is a powerful tool for exploring and creating cata
lytic function in biological macromolecules. This review focuses on ex
amples and current strategies for the application of random mutagenesi
s of target genes coupled with direct selection in vivo. The technolog
y is illustrated by the dissection of catalytic features in chorismate
mutase. Future avenues for identifying and evolving catalytic antibod
ies are also discussed.
