Although conventional ion-exchange HPLC and ion-pair reversed-phase HP
LC using tetrabutylammonium hydroxide allowed ready determination of u
ridine 5'-diphospho-alpha-D-glucose and its hydrolysis products, neith
er method was suitable for the determination of uridine 5'-diphospho-a
lpha-D-glucuronic acid (alpha-UDPGA). However, mixed ion-pair reversed
-phase HPLC, using a combination of tetrabutylammonium hydroxide and t
etraethylammonium hydroxide (1:1), has been found to offer a convenien
t and effective means of simultaneously determining alpha-UDPGA and it
s hydrolysis products, UDP, UMP and cUMP.
