The separation of a model mixture of six indole alkaloids by high-perf
ormance liquid chromatography (HPLC) and high-performance thin-layer c
hromatography (HPTLC) on the normal- and reversed-phase Armsorb suppor
ts, respectively, was studied. A procedure for the analysis of extract
s from plants cell callus culture of Rauwolfia serpentina and hairy ro
ots of R. serpentina and R. vomitoria in the presence of these alkaloi
ds and its derivatives with different chromatographic systems was elab
orated. It was found that the alkaloid compositions of plant cell cult
ures and hairy roots of R. serpentina and R. vomitoria are identical.
With HPLC analysis a higher precision of alkaloid determination in sol
ution and measurements of concentration is achieved. HPTLC analysis gi
ves a qualitative identification of the extracts. The combination of t
hese high-performance methods provides quantitative and qualitative an
alyses of indole alkaloids such as ajmaline, ajmalicine, reserpine, ra
ucaffricine, serpentine and yohimbine.
