To examine the relationship between alpha(1)-antitrypsin (alpha(1)AT)
specific activity and tumor necrosis factor alpha(TNF alpha) concentra
tion in synovial fluid from 48 patients with rheumatoid arthritis. Met
hods.The specific activity of alpha(1)AT was calculated from the measu
rement of alpha(1)AT concentration (by rocket immunoelectrophoresis) a
nd elastase inhibitory capacity. TNF alpha was detected by enzyme-link
ed immunosorbent assay. Results. TNF alpha concentrations correlated w
ith the extent of alpha(1)AT inactivation. Conclusion. Our findings ar
e consistent with a role of elastase in TNF alpha release within the i
nflamed joint.
