Two monoclonal antibodies against the Val928-Lys945 fragment of the al
pha-subunit of pig Na+,K+-ATPase were obtained. The location of their
epitopes was determined inside the cell by the enzyme-linked immunoass
ay method using intact and inverted cells (erythrocytes and pig embryo
kidney cells) as antigens. By means of chemical modification and enzy
matic degradation of the Val928-Lys945 fragment, the epitopes of these
monoclonal antibodies were determined in detail. Based on the obtaine
d data, models of the transmembrane organization of the COOH-terminal
region of Na+,K+-ATPase alpha-subunit were proposed.
