We examined cellular and subcellular localization of endothelial nitri
c oxide synthase by using immunoelectron microscopy. The results showe
d that immunoreaction products were associated with membranous structu
re, likely the endoplasmic reticulum and cytoplasmic vesicles. These m
embranous localization of imunoreactivities at electron microscopic le
vel corresponded to the perinuclear granular structures observed by li
ght microscopic immunohistochemistry for endothelial nitric oxide synt
hase as well as NADPH diaphorase histochemistry.
