A monooxygenase specific for theobromine demethylation was purified fr
om the cell-free extract of Pseudomonas putida No. 352 to homogeneity.
Caffeine appears to be demethylated to 7-methylxanthine at least by t
wo enzymes, caffeine demethylase and theobromine demethylase. The latt
er is sensitive to Zn2+ and possibly caused an accumulation of theobro
mine in P. putida No. 352.
