F. Altepeter et Uk. Posselt, REGENERATION OF FERTILE PLANTS FROM CELL-SUSPENSION-DERIVED SMALL-CELL COLONIES OF LOLIUM-PERENNE (L), Plant breeding, 113(4), 1994, pp. 327-330
A defined method for the rapid establishment of cell suspensions relea
sing small-cell colonies and their regeneration to fertile plants is d
escribed. Cell suspensions were obtained from 10 mature, lengthwise-di
vided embryos. Five of the 10 suspensions regenerated green plants. Fi
ltration of suspensions through a 150 mu m mesh followed by regenerati
on was carried out 28 days after initiation of the suspensions, to dem
onstrate a rapid release of regenerable small cell colonies from cell
suspensions using the protocol described. Two of the 10 filtered suspe
nsions regenerated green plants from small cell colonies.
