A high performance liquid chromatography procedure has been developed
for the assay of a metoclopramide and ondansetron mixture in 0.9% sodi
um chloride injection. The separation and quantitation are achieved on
a base deactivated octylsilane column at ambient temperature using a
mobile phase of 77:23 v/v 0.01 M phosphate buffer, pH 4 - acetonitrile
at a flow rate of 1.0 mL/min with detection of analytes at 273 nm. Th
e separation is achieved within 15-20 min with sensitivity in the ng/m
L range for each analyte. The method showed linearity for metocloprami
de and ondansetron in the 12.5 - 50 and 5-20 mu g/mL ranges, respectiv
ely. Accuracy and precision were in the 1 - 2% and 0.3 - 1.3% ranges,
respectively, for both drugs. The limits of detection for metocloprami
de and ondansetron were 49 and 20 ng/mL, respectively, based on a sign
al to noise ratio of 3 and a 20 mu L injection.
