Abstract
A practical method for the solid-phase synthesis and purification of C -14 labeled oligonucleotides is described. A streamlined procedure for the synthesis of 5'-O-dimethoxytrityl-2-(C-14)-thymidine 3'-(2-cyanoe thyl-N,N-diisopropyl) phosphoramidite from commercially available 2-(C -14) labeled thymidine nucleoside is detailed. Methodology for the man ual synthesis, isolation, and purification of a high specific activity oligonucleotide phosphorothioate 20-mer (ISIS 2105) are elaborated.
