Using a newly developed temperature-gradient gel electrophoresis appar
atus, mutations in the K-ras oncogene and p53 tumor supressor gene wer
e analyzed for single-strand conformation polymorphism (SSCP). The mob
ilities of single-stranded DNAs, carrying various mutations, change -
depending on the gel temperature during electrophoresis. Therefore, te
mperature-gradient single-strand conformation polymorphism (TG-SSCP) a
nalysis can provide useful information concerning the optimum temperat
ure for SSCP and may also be used to screen for various mutations or p
olymorphisms in a single electrophoretic run.
