Methodology for in situ infrared monitoring and analysis of solid-phas
e organic reactions has been developed. The real-time, single-bead cap
abilities of this technique are illustrated through the analysis of so
lvent diffusion into, and subsequent wash-out from, a 70-mu m aminomet
hyl polystyrene bead. This report further illustrates application of t
his technology for solid-phase synthesis with real-time analysis using
two example reactions. Analytical hardware and the inherent benefits
of real-time analysis with respect to precursor referencing and the qu
ality, utility, and interpretation of the infrared data are also discu
ssed.
