Leaf base and mesocotyl explants derived from in vitro-grown seedlings
of Echinochloa colona were cultured on Murashige and Skoog's (MS) med
ium containing various concentrations of benzyladenine (BA), alpha-nap
hthaleneacetic acid (NAA) and kinetin. Leaf base and mesocotyl segment
s exhibited optimal morphogenetic response by using 6.66 mu M BA with
2.68 mu M NAA. Induction of rooting from regenerated shoots was readil
y achieved in half strength MS medium without organics and growth regu
lators. Histological studies revealed the sequence of shoot bud regene
ration in the monocot system. The in vitro-raised plants were establis
hed in chromite minewaste.
