An automated sandwich enzyme-linked immunosorbent assay (ELISA) to mea
sure human immunoglobulin E (IgE) in the concentration range of 0.2 to
100 ng/mL was developed. The ELISA uses a rabbit antihuman IgE for th
e capture antibody, a biotinylated goat antihuman IgE for the defectio
n antibody, and alkaline phosphatase conjugated to streptavidin as the
detection system. Assay precision was improved over manual methods an
d had a coefficient of variation (CV) of 3-8%. Accuracy was confirmed
by measurement of standards calibrated against World Health Organizati
on (WHO) standards. Cross-reactivity against other human immunoglobuli
ns was less than 0.001%. This ELISA successfully measured IgE concentr
ations of culture supernatants from human lymphocytes.
