Shoot proliferation from axillary buds of Prunus mume Sieb. et Zucc. w
as obtained on Woody Plant Medium (WPM) supplemented with 1 to 5 mu M
benzyladenine, 3% sorbitol and solidified with 0.5 to 0.7% agar. Effec
ts of different carbon sources on shoot proliferation were examined. G
lucose provided better shoot proliferation than sucrose, sorbitol and
fructose. In the presence of sucrose, leaf chlorosis occurred and shoo
ts gradually declined. Best rooting percentage was obtained on WPM sup
plemented with 1 mu M naphthaleneacetic acid. Rooted plantlets were ac
climatized under intermittent mist. However, survival rate was relativ
ely low (20 to 30%).
