U. Kucharska et J. Maslowska, COMPARISON OF ENZYMATIC AND CHEMICAL METHODS FOR DETECTING AND DETERMINING TRACE QUANTITIES OF ORGANIC-ACIDS AND AMINO-ACIDS ON CHROMATOGRAMS, Chemia Analityczna, 41(6), 1996, pp. 965-973
Either activation or inhibition of dehydrogenase type enzymes such as
isocitrate dehydrogenase (ICDH), lactate dehydrogenase (LDH), glutamat
e dehydrogenase (GLDH), alcohol dehydrogenase (AIDH) and aldehyde dehy
drogenase (AdDH) has been used to develop new methods for detecting an
d determining trace quantities of selected organic acids and aminoacid
s on chromatograms. It has been found that the developed methods are s
uitable for the determinations of these compounds present in quantitie
s from 0.01 to 10 mu g. The sensitivity was increased by 10-100 times
compared to that of spectral methods. In addition, the proposed enzyma
tic methods are characterized by a good reproducibility, a simple dete
rmination procedure within a short period of time, and readily availab
le equipment. They are particularly-useful in serial determinations.
