Amine oxidase was isolated from eight-day-old Trigonella foenum-graecu
m seedlings grown in the dark, using rivanol lactate and ammonium sulp
hate fractionations, heat denaturation and hydroxylapatite chromatogra
phy. The yellow enzyme is a monomer M(r) of 80 000 with pI 6.8, A(max)
at 280 nm and a shoulder at 415 nm. Substrate specificity and inhibit
or interactions were similar to those of pea amine oxidase. The N-term
inal of the enzyme was blocked and no reaction was observed with antib
ody against amine oxidase either from Escherichia coli or Micrococcus
luteus.
