Abstract
The capability of the cysteine protease ficin to form peptide bonds in frozen aqueous solution is shown. Freezing of the reaction mixture re sulted in increased peptide yields in kinetically controlled coupling of ester substrates with leucine amide. The influence of temperature, concentration of the amino component, pH before freezing and various s olubilizing N-alpha-proctecting groups was studied.
