Different fractions of a mixture of protein-pigment complexes have bee
n separated from one another by non-denaturing gel electrophoresis. Th
ese complexes were prepared for observation by inserting electron micr
oscope grids directly into the focused bands of the pigment-protein co
mplexes and resuming electrophoresis for a brief time, so that the com
plexes were deposited onto the grids. It was found that complexes depo
sited from each band exhibited distinctly different appearances. It wa
s also found that the exact conditions of electrophoretic deposition o
nto the grids affected the appearance of the complexes. The protein-pi
gment complexes were characterized additionally by spectroscopy and de
naturing gel electrophoresis.
