Ten pentadecadeoxynucleotides were used to study several fragments of
18S rRNA. The resulting heteroduplexes were hydrolyzed by RNase H. Onl
y four probes bound to fragments 980-994, 987-1001, 1025-1039, and 103
2-1046 of 18S rRNA. None of the probes inhibited tRNA-dependent bindin
g of oligo(U) to the 40S subunit. The probes complementary to fragment
s 987-1001 and 1025-1039 inhibited translation of poly(U) and formatio
n of the pAUG-40S-Met-tRNA(Met)-elF-2-GTP complex, implying that these
fragments are involved in ribosome-mRNA interaction.
