EXPRESSION OF YEAST O-6-METHYLGUANINE-DNA METHYLTRANSFERASE (MGMT) GENE

We cloned the full genomic DNA of yeast (Saccharomyces cerevisiae) O-6 -methylguanine-DNA methyltransferase (MGMT) gene and examined its expr ession. The expression of yeast MGMT gene is decreased when cells reac h stationary phase and cannot be induced by the pretreatment with alky lating agents, methylmethanesulfonate (MMS) or N-methyl-N'-nitroso-N-n itrosoguanidine (MNNG). The transcription initiation site was determin ed by primer extension analysis. This analysis showed that the authent ic start codon is the ATG at position +32 from transcription initiatio n site.
