A chromogenic test with L-proline p-nitroanilide as a substrate was de
veloped to detect proline iminopeptidase activity from Eikenella corro
dens. Optimal assay conditions for enzyme detection were 50 degrees C
and pH 7.5-8.0. Forty paper point samples from adult periodontitis pat
ients were screened for Eik. corrodens and proline iminopeptidase acti
vity, to determine whether this enzyme is a marker for the pathogen. T
wenty samples were positive for Eik. corrodens, and the levels of enzy
me activity and populations of Eik. corrodens were found to correlate
(P < 0.05).
