Abstract
Site-directed mutagenesis using two different DNA polymerases was comp ared on the Saccharomyces cerevisiae CTA1 gene. It showed different ap plication possibilities of various available DNA polymerases. While th e Thermus aquaticus DNA polymerase can be used for in vitro evolution by random production of mutations, the Thermus brockianus DNA polymera se, under the conditions described, is suitable for directed mutagenes is employed in the studies of structure-function relationships in the encoded proteins.
