Protoplasts of Daucus carota were coated by a hydrophobized polysaccha
ride, such as cholesterol-bearing modified pullulan (CHP) or cholester
ol-bearing xyloglucan phosphate (CHXP), that provided an artificial ce
ll wall to the protoplast. The polysaccharide-coating was confirmed di
rectly by a confocal laser microscopy using FITC-labeled polysaccharid
e derivative. The artificial cell wall certainly increased the viabili
ty of the protoplasts even in the presence of cellulase.
