Ri. Monge et al., PURIFICATION AND STABILIZATION OF PHENYLALANINE AMMONIA-LYASE FROM SPORIDIOBOLUS-PARAROSEUS, Biotechnology techniques, 9(6), 1995, pp. 423-428
The purification of phenylalanine ammonia-lyase (PAL) from S. pararose
us is described: PAL is recovered from cellular extracts by acid preci
pitation of contaminating proteins, nucleic acids and carotenoids; con
centrated by ultrafiltration and purified by ion exchange chromatograp
hy in DEAE-Sephacel, with 80% yield and 32.6 fold purification. Elimin
ation of carotenoids and stabilization with glycerol are introduced in
the methodology to increase stability and purification yield.
