A yeast system based on a S. cerevisiae mutant strain deficient in cel
l integrity, was used to achieve the release of yeast expressed virus-
like particles (VLPs) by simple osmotic shock. Yeast cells, grown in t
he presence of 1M sorbitol, lysed and released the intracellular conte
nt upon being transferred to non-osmotically stabilized medium. The re
lease of VLPs was followed by Western blotting determination of the p1
protein.
