Abstract
A new restriction endonuclease (ENase), Sgfl, has been isolated from t
he bacterium Streptomyces sp. Sgfl recognizes the 8-bp palindrome 5'-G
CGATCGC-3' and cleaves double-stranded DNA after the T in this sequenc
e, producing a two-base 3' overhang compatible with PvuI termini. SgfI
is a rare-cutting ENase and should be useful for megabase mapping exp
eriments.
