Abstract
Twenty different fractions of hematoporphyrin derivatives (HpD) and ei ght fractions of an HpD dimer mixture were isolated utilizing isocrati c reversed-phase ion-pair high-performance liquid chromatography. Thes e fractions were characterized by UV-visible and fluorescence spectrop hotometry. Fluorescence quantum yields and photokill efficiency for ea ch fraction in PTK2 epithelial cells were obtained. Results indicate t hat some part of the photoactivity exhibited by HpD may be due to impu rities present in the HpD starting material, hematoporphyrin-IX dihydr ochloride, depending on its source. It was also found that hematoporph yrin D, a commercial acetylated product formed during synthesis of HpD , contained a higher percentage of monomers than would be expected.
