The design of haptens to generate antibodies with the potential to cat
alyse carbanionic cyclisation reactions is introduced in the context o
f Dieckmann cyclisations and related reactions. Syntheses of five-memb
ered ring haptens containing a sulphone, to represent the transition s
tate(s) associated with the tetrahedral intermediate, and an amino gro
up, to promote the formation of a general base, are described. Results
with partially purified antibodies are reported showing that the targ
et reaction can be catalysed by antibodies.
