In order to prepare an active enzyme extract of UDP-glucose: scopoleti
n glucosyltransferase from suspension cultured cells of Solanaceous sp
ecies, the analytical conditions were improved, extraction of crude en
zyme optimized, and cells of Duboisia myoporoides, Nicotiana plumbagin
ifolia, and Solanum aviculare were compared as sources. Cells of Duboi
sia gave best yields, which could be further increased by precultivati
on in the presence of phenobarbital. For extraction of crude enzyme, T
ris and phosphate buffers were compared, with addition of mercaptoetha
nol, EDTA and ascorbate. The activity of SGTase exhibited 14.10(-3) nK
at. mg(-1) of proteins using 130 mu M of scopoletin as a substrate.
