Abstract
A cis-acting regulatory region within the first intron of the human pl atelet-derived growth factor (PDGF) A-chain gene has been identified t hat functions to negatively regulate transcription of PDGF A-chain pro moter/CAT reporter constructs in both A172 and HeLa cells and that fun ctions independent of position, orientation, and promoter context. Fur ther dissection of this region revealed several independently acting n egative regulatory elements that exhibited cell-type specificity. Thes e results suggest that the first intron of the PDGF A-chain gene conta ins negative regulatory elements that may cooperate to regulate the ce ll-type specific expression of the PDGF A-chain gene.
