A depth filter perfusion system (DFPS), equipped with a 40-mu m polypr
opylene depth filter for cell immobilization, was used for the continu
ous production of tissue plasminogen activator (t-PA) from recombinant
Chinese hamster ovary cells. Final cell density in the DFPS with oxyg
en control was 1.8x10(7) cells/mL of the total working volume and maxi
mum t-PA productivity was 2.63 mg/L/day. Dissolved oxygen concentratio
n in the filter matrix was successfully controlled by air sparging and
stable operation was possible for more than 20 days.
