Confocal fluorescence spectral imaging technique was used to analyse t
he intracellular molecular interactions and distribution of mitoxantro
ne (MTX) within synchronised K562 cancer cells in the S-, G(2)- and M-
phases of cell cycle. The spectral decomposition of the original fluor
escence signal coming from a microvolume has been carried out to calcu
late the intracellular distribution of free MTX and MTX/DNA complex. T
he analysis of the distribution of MTX in different phases of the cycl
e has been applied to study the phase-dependent changes of intracellul
ar molecular interaction of MTX.
