By using a wide-pore (30 nm), microspheric silica (8 mu m) as the matr
ix and gamma-aminopropyltriethoxysilane as the coupling agent, a stati
onary phase of biospecific affinity chromatography was prepared by che
mical binding nicotinamide adenine dinucleotide (NAD) to the gamma-ami
nopropyl modified silica, Using the phosphate buffer (PH = 7.0) as a m
obile phase, the separation of some nucleotides is accomplished by the
detection of UVD (260 nm). The effect of changed ion strengh of eluen
t on the resolution of nucleotide (AMP, ADP and ATP) has been studied.
