Abstract
Thick biological specimens prepared as whole mount cultured cells stai
ned with histochemical reactions, such as thiamine pyrophosphatase, gl
ucose-6-phosphatase, cytochrome oxidase, acid phosphatase, DAB reactio
ns demonstrating specific cell organelles such as Golgi apparatus, end
oplasmic reticulum, mitochondria, lysosomes, peroxisomes and pinocytot
ic vesicles, were observed by ultrahigh voltage electron microscopy at
accelerating voltages of 400-1000 kV producing stereo-pairs. As a res
ult, those cell organelles were observed 3-dimensionally and the relat
ive relationships between these organelles demonstrated.
