A sensitive and specific method has been developed for the simultaneou
s analysis of different O-6-alkylguanines. The cross-reactivity of two
different antibodies raised against O-6-methylguanosine and O-6-butyl
guanosine for a series of O-6-alkylguanines was exploited for the immu
noaffinity purification of biological samples before quantitative anal
ysis by gas chromatography/mass spectrometry. The method can be applie
d to the detection of O-6-alkylguanines in DNA and appears to be usefu
l for studying chemical carcinogen mechanisms in animals and possibly
for the detection of human exposure to alkylating agents.
