This paper reviews existing high-performance liquid chromatographic (H
PLC) methods for the analysis of phosphatidic acid (PA) in various sam
ple matrices. In addition to the introductory background discussion on
important aspects of PA in lipid biochemistry, the review provides co
mprehensive coverage in the areas of derivatization techniques, detect
ion methods, and HPLC separation techniques. Conversions of PA to suit
able derivatives enhance the detection sensitivity and improve the chr
omatographic behavior of the analytes. Detection methods include the u
se of state-of-the-art detectors and are discussed in terms of sensiti
vity, specificity, and compatibility with analytical systems. Pertinen
t normal-phase and reversed-phase HPLC data for PA are compiled from p
ublished methods.
