Antiischemic cardioprotective effects of a new 3-hydroxypyridine antio
xidant mexidol were investigated after experimental coronary artery oc
clusion in rats and dogs. influence of the drug on myocardial infarct
size was estimated in rats by ratio of necrotic to ischemic zone size
in 4 hours after ligation of anterior descending coronary artery. Acti
on of mexidol on collateral coronary blood flow, dp/dt(max), and some
parameters of energy metabolism of ischemic myocardium was studied in
dogs. Necrotic to ischemic zone ratio was lower in mexidol treated (30
mg/kg) than in control animals (46+/-5,6% and 68+/-4,3%, respectively
). Compared to control group, intravenous administration of 5 and 20 m
g/kg of mexidol significantly increased collateral coronary blood flow
and dp/dt(max), decreased excess lactate in the blood drained from th
e ischemic zone and inhibited glucose consumption by ischemic myocardi
um.
