With chlorotetracycline (CTC) and fluphenazine (FFZ) respectively fluo
rescence the localization of (membrane- bound) calcium and calmodulin
was observed. Freshly isolated protoplasts of dihaploid Solanum tubero
sum (DH 81.71463) showed a hight cytoplasmic fluorescence. Later on, w
hen cell division took place, the fluorescence was localized around th
e plasmatic membrane. Dividing cultures gave a strong CTC and FPZ fluo
rescence during the first days of protoplasts cultivation. Cell walls,
nuclei and plastids were without fluorescence according to these fact
s we conclude that no specific distribution of membrane-bound calcium
and activated calmodulin or rearragements of cellular calcium pools ca
n be observed during cultivation of leaf mesophyll protoplasts.
