P. Cognez et al., ELIMINATION OF LINEAR PARAMETERS IN NONLINEAR REGRESSIONS - A FAST AND EFFECTIVE METHOD FOR THE DETERMINATION OF BINDING PARAMETERS, International journal of pharmaceutics, 123(1), 1995, pp. 137-142
A semi-linear least-squares regression methodology is proposed to extr
act binding parameters from experimental binding data. After legitimat
ion of simplification and suitable mathematical transformation, the on
ly non-linear parameter which remains to be searched for by a non-line
ar least-squares regression is the affinity constant. As a result, num
ber of sites and affinity constants are easier to grasp than with a fu
ll non-linear regression. This methodology can be applied to saturatio
n as well as to displacement binding studies. It has been tested with
binding data of QNB and alpha BTX on rat hippocampus synaptosomes.
