Ag. Katrukha et al., A NEW METHOD OF HUMAN CARDIAC TROPONIN-I AND TROPONIN-T PURIFICATION, Biochemistry and molecular biology international, 36(1), 1995, pp. 195-202
New rapid and effective procedure for simultaneous purification of hum
an cardiac troponin I and troponin T has been developed. Affinity chro
matography on immobilized monoclonal antitroponin I antibody C5 was us
ed for purification of the whole troponin complex with a yield of 120
mg from 100 g of tissue. Isolated troponin I and troponin T (about 20
and 35 mg from 100 g tissue) were obtained by conventionally used ion-
exchange chromatography. Antibody C5 recognizes conservative epitope o
f troponin I, therefore the method is applicable for purification of s
keletal and cardiac troponin from a number of different animal species
.
