Domains differing in the degree of evolutionary conservatism were reve
aled within the 17S ribosomal DNA (rDNA) sequence from Tetrahymena pyr
iformis by means of computer analysis. The method proposed allows evol
ution of rDNA structure to be studied in eukaryotes. Superconservative
regions revealed within the DNA sequence tested can be used to constr
uct universal primers for studying rDNA structure in various taxa by m
eans of polymerase chain reaction (PCR).
