In vitro basal and GHRH-induced GH secretion was studied in adult male
rat pituitaries that were either intact, orchidectomized or treated w
ith ethylene dimethane sulphonate (EDS) . Two weeks after orchidectomy
or EDS injection (75 mg/kg) , the animals were decapitated and their
hemipituitaries incubated in Krebs-Ringer-Bicarbonate (KRB) with GHRH
(10(-6) M) . GH concentrations in the medium were measured after 30,60
and 120 min incubation. We found that GHRH always stimulated GH secre
tion and that orchidectomy and EDS treatment reduced basal and GHRH-in
duced GH secretion in a similar way.
