To obtain a low cost, beaded chromatographic matrix, calcium alginate
beads were cross-liked with epichlorohydrin, and calcium was removed b
y sodium citrate treatment. The cross-linking reaction for obtaining s
table beads was optimized. For purification of haemoglobin by ion exch
ange, cross-linked calcium-free alginate beads have a q(max) of 60 mg/
ml and a K(d) of 0.02 mg/ml gel, while for affinity polygalacturonase
purification K(aff) was 0.007 ml/g.
