A simple method of generating pH gradients for use in capillary isoele
ctric focusing was described and evaluated experimentally. In the prop
osed scheme, the cone shaped capillary filled with a conventional buff
er is used to produce an effect of nonuniform heating which then gener
ate temperature and corresponding pH gradients along the capillary axi
s. Concentration and separation of proteins by applying a simple isoel
ectric focusing system based on this principle is demonstrated. An ine
xpensive absorption imaging detection system is used to continuously m
onitor progress of the separation and concentration processes.
