A modification of the activation methodology of hydroxylic support usi
ng p-toluensulfonyl chloride is described. The methodology is applied
to the corn cob. The influence of several treatments of the support an
d of the presence of pyridine or ethanolamine in the activation proces
s is discussed. Endonuclease Staphylococcus aureus and pancreatic ribo
nuclease are immobilized by this methodology. The enzymatic derivative
s are active in the hydrolysis of RNA and DNA (only endonuclease S. au
reus).
