Using glucoamylase, alpha-galactosidase, and alpha-glucosidase of the
micellar fungus Asp. awamori as examples of secreted glycoproteins, th
e relationship between glycosylation and secretion was studied using t
unicamycin and 2-deoxy-D-glucose as inhibitors of N- and O-glycosylati
on, respectively. Tunicamycin suppresses N-glycosylation and thus decr
eases the level of secretion of all four glycosidases. 2-Deoxy-D-gluco
se, which predominantly suppresses O-glycosylation, has no effect on t
he secretion level.
