Medium components that may influence the emergence of adventitious roo
t primordia in rhizome buds of Lapageria rosea (Ruiz et Pav.) cv. Nash
court in vitro were examined. Sucrose was the only medium component te
sted that affected root emergence. Increasing the sucrose concentratio
n in the medium from 10 to 30 g1(-1) stimulated adventitious root emer
gence from rhizome buds that had been proliferated on a medium contain
ing the gibberellin biosynthesis inhibitor paclobutrazol (5 mu M). Suc
cessful clonal propagation of L. rosea cv. Nashcourt can now be achiev
ed by a rhizome bud proliferation stage(s) in the presence of paclobut
razol followed by an adventitious root emergence stage in the absence
of paclobutrazol.
