Methods were tested for the preparation of monospecific antisera again
st buffalo IgG1 and IgG2. The heterologous antisera raised in guinea p
igs and rabbits showed extensive cross-reaction with the other subclas
s. The cross-reacting antibodies were removed either by using immunoad
sorbent or by addition of soluble protein of reciprocal subclass. The
pre- and post- absorption antisera were tested for relative potency, s
pecificity and degree of cross-reactivity by immunodiffusion, immunoel
ectrophoresis and reverse radial immunodiffusion tests. The monospecif
ic antisera obtained after addition of soluble proteins were better th
an those obtained after adsorption with immunoadsorbent.
