The analysis of free amino acids in mammalian cell culture media can g
ive valuable information on the metabolism of particular commercially
valuable cell lines. Analysis of spent culture media indicates potenti
al limiting nutrients. However, analysis over the whole culture period
enables a kinetic approach to allow understanding of amino acid uptak
e and potential strategies to improve media design. This communication
details the use of a less commonly used HPLC protocol, indicating var
ious advantages and disadvantages. Further, the batch kinetics of amin
o acid metabolism of two cell lines are discussed.
